This Lab-Aids DNA Staining Kit provides an engaging science education for intermediate and junior high science students. The experiment is based on the Feulgen reaction where DNA in plant cells are stained purple for easy viewing. Your students macerate, stain and mount specially grown root tips (provided in the kit), and then locate and diagram DNA in the cells.
Finally, the students are asked to observe and draw the various stages of cell division. This enables the students to develop a better understanding of the hereditary material in plant chromosomes. All aspects of the lab activity are performed by the individual student.

DNA staining using the Feulgen reaction is a technique used in histology to identify chromosomal material or DNA in cell specimens. It depends on acid hydrolysis of DNA, therefore fixating agents using strong acids should be avoided. In school, science students normally perform DNA staining experiments in Biology class in order to gain basic understanding on the various stages of cell divisions.
The Feulgen reaction proposed by Feulgen and Rossenbeck 75 years ago is one of the cytohistochemical reactions most widely used in biology and medicine. It allows DNA in situ to be specifically stained based on the reaction of Schiff or Schiff-like reagents with aldehyde groups engendered in the deoxyribose molecules by HCl hydrolysis.
